Oligos are synthesized via solid-phase synthesis method and find applications in various fields, including the PCR tests for viruses and pathogens detection, DNA sequencing artificial gene synthesis, rare diseases therapeutics, vaccines and frontier biomedical solutions. However, before these oligonucleotides can be used in downstream applications, they must be cleaved from the solid support and have their protecting groups removed.

AUTOMATED LOADING/ DRAINING STATION

CLEAVAGE AND DEPROTECTION INTO A SINGLE STEP

At the completion of the synthesis process, the oligos have to be removed from the solid support and subsequently deprotected. These manual steps have a direct impact on the final yield, as well as on the productivity and on the lab’s workflow, due to the fact that their completion takes several hours. These steps are usually carried out by immersing the plates into ammonia solutions or other reagents, thus requiring an evaporation step and, therefore, creating unpleasant lab conditions.

In wavePREP, liquid reagents turn into gas, cleaving and deprotecting oligos simultaneously. Reaction conditions are accurately controlled to ensure complete process control and to maximize the yield. Once the procedure has finished, oligonucleotides can be directly desalted, for example with acetonitrile, and eluted from the solid support with the appropriate buffer solution without further steps.

Principle of operation

WavePREP offers a single-step approach to cleavage and deprotection, eliminating the need for evaporation. The reaction occurs by heating the cleavage and deprotection reagents using microwave technology, causing its transition to the gas phase. Precise control over the temperature and pressure of both the vapor and liquid phases ensures optimal yield and safe operation. Consequently, at the end of the process, there is no requirement for evaporation, and the oligonucleotides can be directly eluted for following work-up.
